New vectors for color deconvolution

Hey everyone,
So I have eosin-only stained tissue and I need to be able to create new eosin vectors so that I can use it for an H&E color deconvolution. I am going to be getting hematoxylin-only images soon. I ran into some blogs explaining it using imagej but I really need to a way to use matlab. Somehow have the images be read in and then it pop out a vector. Any help or direction would be greatly appreciated.

See this demo: http://www.mathworks.com/products/demos/image/color_seg_k/ipexhistology.html which uses k-means to do color segmentation.
