How can I work out the coordinates of a point on a DICOM image based on grayscale?

I need to work out the coordinates of a certain point (it is a white circle surrounded by black square which in turn is surrounded by white square) on a dicom image. I think I need to use a region of interest, maybe roicolor but not really sure where to go from there. Help will be much appreciated.
