how to measure the threshold value? i need a method or function that help me to measure the threshold value to use it in a wavelet_based image segmentation (my image is the gray scale image)

You don't measure it - you specify it. So you don't have it somehow and need to measure it to discover what it is, you need to specify in advance what it is so you definitely will know what threshold you used. Please start your own discussion thread, and post your images.

It's the one that works best for you - the one that divides foreground and background in a way that is most useful to you. There is no one algorithm that is best for all images ever captured in the world.

A good place to start if you want to find bright bits, is halfway between the mean (or mode or median) of the image, and the maximum of the image.
