Finding error as the distance between the centroid of two blobs in an image

The yellow square is a plane on which light is reflected from a square mirror. First, I would like to find the centroid of the yellow square and then find the centroid of the white polygon and finally calculate the distance between the two centroids. Can anyone help me with regard to this? Thanks you in advance.
