Doubts about image processing

Hello,
I am doing a research where i have to work with images from the corneal nerves, and they are really dark. These pictures where taken by a confocal microscopy. I am having difficulties to divide the pictures in blocks. I really need to do this because i want to apply the histogram to small regions and then do the segmentation. Can anyone help me?

Could you explain the difficulties you have with any details? What exactly means "really dark" - are they completely black? What kind of images are you working with? Can you post a small example of the images and the code?

the images that i am working is something like this http://www.iovs.org/content/48/5/2017/F1.small.gif.
I want to isolate the nerves (the white stripes). I am using filters like the median, the functions imadjust, adapthisteq, bwmorph, rangefilt...
I can't show you my code because it is really confusing.

I would recommend using photoshop (or equivalent) first to edit the image to a usable format, as photoshop in that matter is more user-friendly, though less scientific.
