Any sample to detest a sample template picture from a big picture

The sample template will exactly match the portion of the big picture.
Which example or toolbox should start with.

There are a few ways to go really. Certainly the Computer Vision System Toolbox has a number of possibilities for that:

You can do it with normxcorr2() in the Image Processing Toolbox. Run this demo:
