Your terminology is confusing.

A .mat file is a data file.
You can read the file into the MATLAB workspace using the load function, or into Simulink using the From File block.

There are many ways to load data from the MATLAB Workspace into Simulink.
One of them is the From Workspace block.
Some other ways are discussed at http://www.goddardconsulting.ca/simulink-interfacing-with-matlab.html#InputData
