segmentation and calculating mean intensity of image

Hi you all guys,

In picture below, i had a matrix at first and by using code "imagesc" i generated that picture. Now i want to calculate mean intensity of Circular Shape. I would be pleased if you could help me out to find its intensity.

i'm trying to find intensity (pixel value) of cysts (the circular shapes)

but i can't find an adaptive method.
