Concatenating element- what the different?

What is the different between those code:
NewVector=[Vector1,element,vector2];
and
NewVector=[Vector1:element:vector2];

E.g.,
[1,3,10] is a three element vector with the elements 1, 3, and 10.
[1:3:10] uses the colon operator and is a four element vector starting with 1 and counting by 3 until 10 is reached or exceeded, so it has the elements 1, 4, 7, 10.
