How to relate an image to diffuse reflectance?

How would I get an rgb color coded image and determine reflectance of that image?

This is a lot more complicated question than you realize. So let's just start with you telling us what you mean by diffuse reflectance. And why do you want to do it with an RGB image instead of a spectrophotometer?
