I have a problem, as your program gives five different pictures in single mat file, suppose 5 pictures of (1200x900 uint16) resolution, then i want (1200x900x5 uint16) matfile. Please help, thank you.

save('CombinedPic.mat', 'combined_pic')

However, if you are reading .jpg files then it is rather unlikely that they are grayscale 16 bit files. Not impossible, but in my years I have only ever seen one actual grayscale .jpg file: everything else has been RGB .jpg files that happen to only use shades of gray.
